EZ-Yeast Transformation Kit
Simple and Fast with Minimal Hands-On Time

The EZ-Yeast Transformation kit is designed for high throughput transformation. It is ideal for cases where large numbers of transformations are required but only a few transformants are needed. Thus, the EZ-Transformation kit is ideal for analyzing "putative positives" obtained from two-hybrid screens. This kit is not designed for high efficiency transformation. However, it is capable of achieving transformation efficiency of up to 1000 colonies per µg of DNA. (For high efficiency transformation use the Alkali Cation Yeast Transformation Kit or the Yeast Spheroplast Transformation kit.

The EZ-Yeast Transformation Kit does not require fully competent cells prior to transformation. Simply add vector DNA to 0.5 ml of overnight yeast culture, incubate overnight at room temperature and plate on selective media (See also The Yeast RPM Kit for isolation of plasmid DNA from yeast for PCR and bacterial transformation, )

• Excellent in cases where only a few transformants are needed, i.e. transformation with a single homogenous vector.
• Transformation efficiency as high as 1x103 per µg DNA.

The protocol in brief:

1. Pellet cells from 0.5 ml of a yeast culture.
2. Add vector and carrier DNA, EZ-Transformation Solution and mix.
3. Incubate overnight at room temperature.
4. Plate the cells the following day on selective plates.
5. Transformant colonies are observed after 2-3 days.
