Yeast Transformation Kit



The Alkali-Cation Yeast Transformation Kit provides all of the reagents required to perform an easy and quick non-electrical method for transforming yeast with linear or plasmid DNA. Host cells are made competent by treatment with a Lithium/Cesium Acetate mixture. The entire procedure may be completed in less than 90 minutes and routinely provides a transformation efficiency of greater than 104 transformants per µg of DNA, using YEP24 plasmid DNA.

The protocol in brief:

  1. Suspend a washed log phase yeast cell pellet in lithium cesium acetate.
  2. After 30 minutes, the competent cells are washed and resuspended in TE.
  3. The competent cells are transformed by adding carrier DNA, histamine, and plasmid.
  4. After 15 minutes PEG is added and 10 minutes later the cells are heat shocked.
  5. The transformed cells are resuspended in SOS media, and after 30 minutes incubation at 30°C (to allow for expression of introduced sequences before selective pressure is applied), the cells are plated on appropriate selective media. Transformants appear in 2-4 days.
