"Research Grade" Plasmid DNA is isolated by a proprietary procedure that is similar to the method used to isolate Cloning Grade Plasmid DNA but also includes several additional steps that minimize endotoxin, RNA, protein and other trace contaminants. An audit trail provides for sample traceability. The isolation procedure starts with a series of hyper- and hypotonic cell washes that strip away a large proportion of the lipopolysaccharide-containing outer membrane. The cells are lysed by conventional methods and precipitated by various procedures to decrease contaminants. After DNA is purified over a matrix column it is post-treated to decrease endotoxin to levels desired. Each post-treatment cycle eliminates approximately 90% of remaining lipopolysaccharides. Purified DNA is measured for endotoxin levels by a standard quantitative LAL test; RNA and protein levels are also examined. Research Grade R.1 Plasmid DNA contains no measurable protein or RNA and has an average endotoxin level of 0.1 EU/µg. Research Grade R.03 Plasmid DNA contains no measurable protein or RNA and has an average endotoxin level of 0.03 EU/µg.
