Rapid Isolation of poly(A⁺)mRNA

This kit is designed for the rapid isolation of mRNA from cells or tissues. The kit allows for the isolation of poly(A+) mRNA directly from a cell lysate or from a total RNA prep. Direct isolation of mRNA from a lysate is very rapid and ideal when multiple samples are needed for RT-PCR of low abundance in RNA and the requirement for maximum mRNA yield is not necessary. Direct mRNA isolation from cell lysates makes it possible to visualize amplified low abundance messages on a gel. Such an amplification from total RNA is not possible. Poly (A+) mRNA yield can be increased 5-10 fold by isolating mRNA from a total RNA preparation. This kit includes a Matrix Collector Rack which allows the Oligo dT30 Matrix particles that bind the RNA, to be drawn onto the side of a microcentrifuge tube. This facilitates removal of unbound, non-polyadenylated RNA and washing of the matrix/mRNA complex.

To isolate mRNA:

1. Oligo dT30 Matrix in Bind Solution A is added to a freshly prepared lysate or total RNA solution and incu- bated 10-30 minutes to allow binding of mRNA to Oligo dT30 Matrix.
2. Remove the bound mRNA from unbound RNA by pulling the matrix to the side of tube with the Matrix Collector Rack and removing supernatant.
3. Wash mRNA/matrix three times by resuspension in Salt Bind B solution and removal of supernatant.
4. Elute mRNA from washed oligo dT Matrix using RNase-free Elution solution.

Yield of poly(A+) mRNA is approximately 1-2% of RNA present in a total RNA Prep. Yield is decreased 5-10 fold when isolating directly from a cell lysate.
