EZ-Yeast Transformation Kit

The EZ-Yeast Transformation Kit is designed for high throughput transformation. It is ideal for cases where hundreds of transformations are required and only several transformants are needed. It is capable of achieving transformation efficiency of up to 1000 colonies per µg of DNA (depending on strain of yeast). The EZ-Yeast Transformation Kit does not require making cells competent prior to transformation.

Detailed EZ-Yeast Transformation Protocol

  1. Grow 1-5 ml of yeast cells either in rich media (YPD) or minimal dropout media.


  2. Transfer 500 µl to a sterile 1.5 ml centrifuge tube; spin 10 sec. and decant supernate.


  3. Add 1-5 µg of vector DNA, 5 µl carrier DNA, 500 µl EZ-Transformation Solution and mix by vortexing at a low to intermediate speed.


  4. Incubate overnight at room temperature.


  5. Plate 50-150 µl of the mixture taken from the bottom of the tube (cells will settle after overnight incubation) on selective plates and incubate at 30°C until colonies are observed (2-3 days).

Note: Different yeast strains give different transformation efficiencies. If poor transformation results are obtained, use cells that are actively growing (OD600 < 2).
