SPIN Protocol

Protocol Outlines
Protocol A: Rapid Isolation of 0.2-300 kb DNA from any Type of
Agarose.
Protocol Time: 15 minutes.
- Melt gel in 400 µl of GC SPIN GLASSMILK in GC SPIN Filter.
- Wash with GC SPIN NEW Wash*.
- Elute DNA with GC SPIN Elution Solution.
Protocol B: Rapid Purification of 0.2-300 kb DNA from
Solution.
Protocol Time: 10 minutes.
For elimination of salts, proteins/enzymes, oligomers, linkers, primers,
traces of detergents or organic solvents.
- Add DNA solution to 400 µl of GC SPIN GLASSMILK in GC SPIN
Filter.
- Wash with GC SPIN NEW Wash*.
- Elute DNA with GC SPIN Elution Solution.
| *Be sure to add 100% ethanol before initial use. Add 2.5, 25, 50,
150 ml for 5, 50, 100, & 150 prep kits |
Protocols
Protocol A: Rapid Isolation of 0.2-300 kb DNA from any Type of
Agarose.
Protocol Time: 15 minutes.
- Melt gel in 400 µl of GC SPIN GLASSMILK in GC SPIN Filter.
- Shake to mix GC SPIN GLASSMILK and add 400 µl to GC SPIN
Filter
- Add gel slice (300 mg maximum/filter).
- Heat to 55°C for 5 minutes to melt gel;
flick tube to mix. Invert every minute to prevent settling of
matrix.
- Spin liquid out of filter into catch tube. Empty catch tube as
needed.
- Wash with GC SPIN NEW Wash (add ethanol to Wash before first use).
- Add 500 µl of GC SPIN NEW Wash to the filter.
- Spin for 30 seconds or until SPIN Filter is emptied of wash.
- Optional: Repeat wash.
- Empty catch tube and spin for 1 minute to "dry" pellet.
- Elute DNA with GC SPIN Elution Solution.
- Transfer SPIN Filter to a fresh catch tube.
- Add 10 to 25 µl GC SPIN Elution Solution and resuspend
GLASSMILK by flicking the tube or by gently vortexing for 1-2
seconds.
- Spin for 30 seconds to transfer eluted DNA to Catch Tube.
| Note: A second elution can increase yield an
additional 10-15% |
- Discard SPIN Filter and cap the tube. DNA in solution is ready
to use without further manipulation.
Protocol B: Rapid Purification of 0.2-300 kb DNA from
Solution.
Protocol Time: 10 minutes.
For elimination of salts, proteins/enzymes, oligomers, linkers, primers,
traces of detergents or organic solvents.
- Add DNA solution to 400 µl of GC SPIN GLASSMILK in SPIN Filter.
- Shake to mix GC SPIN GLASSMILK and add 400 µl to SPIN Filter.
- Add DNA solution (300 µl maximum/filter). Incubate at
room temperature for 5 minutes. Invert tube every minute to
prevent settling of GLASSMILK
- Spin liquid out of SPIN Filter into catch tube. Empty catch
tube Protocol Index as needed.
- Wash with Geneclean SPIN NEW Wash (add ethanol to Wash before first use).
- Add 500 µl of Geneclean SPIN NEW Wash to the SPIN Filter.
- Spin for 30 seconds or until SPIN Filter is emptied of wash.
- Optional: Repeat wash.
- Empty Catch Tube and spin for 1 minute to "dry" pellet.
- Elute DNA with Geneclean SPIN Elution Solution.
- Transfer SPIN Filter to a catch tube.
- Add 10 to 25 µl Geneclean SPIN Elution Solution and
resuspend GLASSMILK by flicking the tube or by gently vortexing
for 1-2 seconds.
- Spin for 30 seconds to transfer eluted DNA to catch tube. A second
elution can increase yield an additional 10-15%.
- Discard SPIN Filter and cap the tube. DNA in solution is ready
to use without further manipulation.
