The p123T vector provides a flexible system for the direct cloning of PCR products. Based on pBluescript II SK+, it allows the cloning of PCR products via both single and multiple dT overhangs. Utilizing the unique series of restriction sites, the same vector can be cleaved by different enzymes (Xcm I, Bcg I and Pfl I) to produce one, two, or three dT overhangs, respectively. This permits either the direct cloning of PCR products (via a single dA extension) or the option of poly-adenylating the PCR fragment and cloning, with more efficiency, via a multiple dA extension. thus less amplified DNA is required. A Sma I site is present to reduce vector background. Msc I sites flank the insert for optimal excision. Blue/white selection by a-complementation is possible.
