Yeast Cell Lysis Kit
To prepare yeast cells for lysis prior to use of the G-NOME DNA Isolation Kit

This kit allows the use of the GNOME^® DNA Isolation Kit for optional isolation of yeast DNA. The yeast cell wall is digested by an enzyme resulting in partial spheroplast formation. The partial spheroplasts can then be lysed by the Cell Lysis/Denaturing Solution contained in the GNOME^® DNA Isolation Kit. [Even though some spheroplasts are made, they are not suitable for transformation. The Spheroplast Transformation Kit utilizes a similar, but more controlled, protocol to make yeast spheroplasts that can be transformed at high efficiency.]

The protocol, briefly, is as follows:

1. Pellet yeast by centrifugation from 100 ml of culture.
2. Resuspend pellet in 1 ml of Yeast Suspension Buffer.
3. Centrifuge, and resuspend pellet in 0.5 ml Yeast Enzyme Enhancer.
4. Add Yeast Enzyme Salts and Spheroplasting Enzyme Mixx; incubate at 37°C for 15-30 minutes and pellet cells.
5. Resuspend pellet in Cell Suspension Solution and continue with the G-NOME^® DNA Isolation Kit protocol.
