ssPHAGE SPIN Kits



These kits contain all the necessary reagents for isolation of highly purified ssDNA from phage in one hour. The process is especially appropriate for rapid preparation of "long read" sequencing templates from ssDNA phage vectors. No enzymatic reactions or organic extractions are required in this protocol.

The kits contain all the necessary reagents and materials for 60, 120, and 600 minipreps. The miniprep protocol is summarized as follows:

  1. Stab phage plaque and inoculate 1.5 ml media in a small culture tube. Grow phage for 8-16 hours.
  2. Spin down 1.5 ml of the bacterial culture and remove 1 ml of supernatant.
  3. Precipitate and pellet phage with ssPhage Drop Buffer.
  4. By resuspending the phage pellet in ssPhage




ssDNA isolated by the ssPhage DNA SPIN Kit: Different sized inserts were cloned in an M13 vector and transformed into host cells. Plaques were picked at random and grown in culture overnight. One ml of each of the culture supernatants was treated according to the ssPhage DNA SPIN protocol and an aliquot of the eluted ssDNA was electrophoresed on a 1% agarose gel. The bands indicate that several different sizes of inserts were cloned.
