LambdaQuick!^® SPIN Kits

This kit is designed for the rapid isolation and purification of l phage DNA from liquid or plate lysates. Because of the need to obtain high titer lysates for optimum DNA yield, methods and growth media necessary to obtain high titer plate lysates are included in the Kit. No CsCl centrifugation is required. The phage are lysed with guanidine isothiocyanate and DNA is bound to a matrix. Typically, 20 µg of l DNA, free of bacterial DNA and RNA, can be isolated per optimized plate lysate prep in one hour.

The protocol is briefly described as follows:

1. Phage plaques in top agarose are removed from plate(s) and centrifuged to separate phage particles; the agarose pellet is washed with LambdaQuick!^® Suspension Buffer and recentrifuged to remove remaining phage.
2. The phage are concentrated by precipitation with ispropanol and resuspended in TE.
3. Phage are incubated with Nuclease Mixx to eliminate cellular DNA and RNA and reprecipitated with Drop Buffer.
4. Phage particles are lysed and the DNA purified by binding, washing, and eluting from Spin Matrix. The DNA is readily cut with restriction enzymes.
