RPM® 4G
Mini MonsterPrep Kit



The RPM® 4G Kit provides the reagents necessary for purification of 3-6 mg of high copy number plasmid DNA in about two hours from 4 g of cells (1/3-1 liter), without the use of organic extractions.

The main elements of the 4G protocol are as follows:

  1. Special osmotic cell wash reduces endotoxin in final DNA prep.
  2. Collection, suspension, and alkaline lysis of bacteria, followed by neutralization and RNase treatment.
  3. Binding of plasmid DNA to GLASSMILK® in the presence of a guanidine chaotrope; elimination of contaminants by washing with ethanol and acetone mixtures.
  4. Elution of purified plasmid DNA from the GLASSMILK® and concentration by precipitation with salt and alcohol.

The RPM® 4G Kit contains protocols specific for high or low copy plasmid/host systems which include unique procedures for eliminating host chromosomal DNA.
