G-NOME® Mini MonsterPrep Kit



The G-NOME® kit was developed by BIO 101 as a rapid and efficient method for isolation of high molecular weight genomic DNA from bacteria, yeast, and animal cells and tissues of all types. No organic extractions are required, and up to 100 µg of DNA can be isolated per prep.

The protocol, briefly, is as follows:

  1. Suspension of tissue or cells (homogenized if necessary).
  2. Lysis of cells in the presence of RNase.
  3. Incubation with protease.
  4. Precipitation of digested proteins and other debris by proprietary "salting out" procedure, followed by centrifugation.
  5. Addition of ethanol to supernatant, followed by isolation of precipitated DNA by spooling or centrifugation.

The kits come in three sizes, suitable for 10, 25, or 100 preparations. Each preparation starts with approximately 100 mg of cells or tissue and yields up to 100 µg of high MW genomic DNA.

From the left lane of each DNA type is uncut, Hind III, and Mbo I cut. JM101 is: uncut, and cut with isoschizimers Mbo I, Dpn I and Sau3A, left to right. Mbo I does not cut DAM methylated JM101. DNAs were cut to completion for 40 minutes at 37°C and separated on an 0.7% Agarose gel.
