GENECLEAN®
For Purification of DNA (200 bp - 20 kb) from TAE Buffered Agarose Gels and Solutions.


Introduced by BIO 101 in 1986, GENECLEAN® was the first commercial product to make use of the rapid, quantitative, and specific binding of DNA by GLASSMILK®. It can be used either to clean-up DNA from solution or to purify gel bands. Although most commonly used for purification of DNA from gels and solutions, the GENECLEAN® Kit has been used to "clean-up" DNA before and after virtually all possible reactions. The entire process takes just 15-20 minutes for gel bands, and less than ten minutes for solutions. Recovery of DNA from solutions typically exceeds 90%, while that from gels is 60-80%. No syringes, slow gravity columns, or alcohol precipitations are involved.

The GENECLEAN® protocol is described in detail in the Protocol section of this catalog. Briefly summarized, it involves just four steps:

  1. Add NaI to the solution* or to excised gel** band; in the latter case, allow several minutes for the gel to dissolve.
  2. Add GLASSMILK® and allow DNA to bind (5 minutes).
  3. Wash away contaminants by sedimentation or by spin filtration in a microcentrifuge (5 seconds or less).
  4. Elute DNA from GLASSMILK® with water or TE (5 minutes).
  5. The GENECLEAN® Kit contains sufficient reagents for 200 purifications.

DNA purified from agarose and solution with the GENECLEAN® Kit. Control consisted of two DNA bands and a smaller diffused band of RNA. When control was purified (in duplicate) from solution, the two DNA bands were recovered and the RNA eliminated. The figure shows recovery of specific DNA bands from agarose; after excising the upper band (in duplicate) and purifying the DNA from the agarose with GENECLEAN®, the eluted DNA was run on a gel.
