This kit is designed for isolation of DNA from bone, preserved tissue, soil organisms, animal by-products, etc. Up to 20 µg of DNA can be purified from each preparation. Special attention has been paid to the production of the reagents of this kit in order to minimize the possibility of contamination by contemporary DNA of extraneous sources. All solutions have been prepared from ultrapure reagents and dispensed in a dedicated laminar flow hood. Relative to DNA isolation kits or reagents that have not been treated in this manner, this kit is DNase-free. No PCR template can be detected with probes to non- human genes. Occasionally, however, traces of human DNA can be detected by multiple, high-cycle PCR. Therefore, this kit is not certified to be free of all DNA traces and it is recommended that proper controls are used.

A brief outline of the protocol:

1. Incubation of homogenized sample in DNA DeHybernation solution.
2. Centrifugation to remove debris.
3. Addition of Ancient DNA GLASSMILK^® to supernatant in Spin Filter.
4. Washing of DNA-matrix complex with three separate wash solutions.
5. Elution of DNA from Ancient DNA GLASSMILK^® in DNA-Free Elution Solution.

Samples of Bison bison bone obtained from the Glenrock Archaeological site¬ (Central, WY - 1670AD) and from the Husdon-Meng (NW Nebraska - 10,000BC), were extracted using the Geneclean for Ancient DNA Kit. The resulting samples were PCR amplified and run on an agarose gel. The samples are loaded in lanes 1-5; lane 6 is an extraction-negative control and lane 7 is a PCR negative control. (Courtesy of K.
