Because of the wide variety of plant tissue types and cytoplasmic non-DNA components, the most consistent results are obtained by first isolating nuclei and discarding non-nuclear fractions. Purified nuclei are lysed to obtain DNA free of phenolics, polysaccharides, and other contaminants that are often difficult to eliminate from DNA preps of plant cells and can inhibit subsequent enzymatic manipulation of the DNA. FLORACLEAN is a modification of the popular G-Nome DNA Isolation Kit that has been designed specifically for isolation of nuclear chromosomal DNA from plant tissues. No organic extractions are required, and up to 400 µg of DNA can be isolated from 1.5 g of tissue in 4-6 hours (more than half of which is incubation time). The isolated DNA is suitable for PCR or enzymatic manipulations.
