Band Cleanup System

One of the more time-consuming procedures in the preparation of cesium-purified DNA is the removal of ethidium bromide and cesium chloride from the isolated density gradient bands. This is conventionally achieved by several organic extractions, followed by overnight dialysis and finally ethanol precipitation. The entire clean-up procedure can add 1-2 days to the already lengthy process of preparing plasmid by this method.

BIO 101's CsCl EZ Band Cleanup System eliminates this lengthy and tedious process entirely. Using this kit, isolated CsCl gradient bands can be completely freed of ethidium bromide and salts in 30 minutes. When employed in conjunction with our simplified CsCl CirclePrep^® Step Gradient Kit, the result is a significant reduction of time and effort to produce supercoiled plasmid DNA. The entire procedure, beginning with a cell pellet to the purified DNA, can be completed in one day - two hours of hands on time and an overnight spin.

The EZ Band Cleanup protocol in brief:

1. Remove DNA band from CsCl gradient with syringe, and load onto cartridge (15 minutes).
2. Wash column with EtBr Washout solution followed by EtOH to remove EtBr and CsCl (10 minutes).
3. Elute with water or TE (5 minutes).
