For preparation of plasmid DNA of the highest purity for research uses, banding in a cesium chloride gradient has long been considered the method of choice. Conventional CsCl-based preps, however, require several days. BIO 101's CsCl Circleprep® Step Gradient Kit allows the entire procedure to be completed in one day.
After a one hour alkaline lysis procedure, the cleared lysate is put in a CsCl step gradient. Following an overnight centrifugation at 45,000 rpm, the purified band of plasmid DNA is removed and purified through a syringe cartridge purification system (see next product page, EZ Band Cleanup System) to remove CsCl and ethidium bromide. After elution from the cartridge, the purified DNA is precipitated and dissolved at the desired concentration for immediate use.
