This media was formulated to do two things: grow host cells to the highest density possible without aeration, and produce high yields of plasmid DNA. GS96 Broth is used in the front end of high throughput automated systems for preparing DNA sequencing templates in 96 well formats.
Typical results of plasmid yield from shaking and non-shaking cultures of GS96 Broth, LB Medium and CIRCLEGROW (duplicate 1.5 ml cultures in capped 1.9 ml tubes) are shown below:

The results show, in a non-shaking 1.5 ml culture (with little airspace in a capped tube) that approximately 3 mg of sequencing template was produced by GS96 Broth. Under these conditions LB medium or CIRCLEGROW produce 1/3 the quantity of plasmid regardless as to whether the cultures were aerated by shaking or not.
